"The investigation is still in progress and an arrest has not yet been made", said Dermot Shea, NYPD chief of detectives, who will be hosting a press conference at One Police Plaza Press Room to discuss this case.
Comello has residences in several places, including Staten Island, Shea said.
Mr Shea declined to say if Mr Comello had a criminal record, but said he had "crossed paths" with the NYPD. He is purportedly a person of interest in the shooting and killing of Frank Cali, 53, in the Todt Hill neighborhood of Staten Island last Wednesday.
At this point, the motive is still to be determined.
"There are multiple, multiple angles that we are exploring", Shea said at a news conference at police headquarters.
CNN is trying to reach Comello's lawyer for comment.
Gambino godfather Paul Castellano was ambushed outside Sparks Steakhouse in December 1985, paving the way for John Gotti to ascend the organized crime ladder to the position of dapper don.
A panicked family member called 911, telling a dispatcher Cali was run over before he was shot, though investigators were still looking into whether the gunman mowed the mobster down, sources said.
When Cali tried to take cover behind his auto, the pickup truck drove into it and "rocked" it significantly, Shea said. Cali was taken to a hospital where he was pronounced dead.
It's "quite possible" that the incident was staged to draw Cali outside and into a confrontation with the suspected shooter, Shea said.
Shea said police have recovered the truck but have not recovered the gun used in the murder.
Federal prosecutors in Brooklyn had referred to Cali in court filings in recent years as the underboss of the Gambino organization, related through marriage to the Inzerillo clan in the Sicilian Mafia.
Authorities alleged Cali and others arranged, through force and threat of force, to receive cash payments from someone who had worked on the project. He was sentenced to 16 months behind bars and was released in 2009.
Gotti died in prison of cancer in 2002.
